A production batch should not be approved simply because a supplier has provided a certificate of analysis or because the material has been purchased before. A change in source, manufacturing site, specification, packaging, transport conditions, or even documentation format can affect the suitability of pharmaceutical raw materials. The practical decision is to approve a material only after the material, supplier, and incoming batch have each passed defined qualification controls.
For quality and safety teams, the process should confirm five points: the material is correctly identified, meets the approved specification, is produced under an acceptable quality system, can be traced through the supply chain, and presents no unmanaged contamination or handling risk. Approval should be documented before the material is released for production use.
Qualification depth should reflect how the raw material will be used. An active ingredient, sterile-process input, high-potency compound, or material with known impurity concerns generally requires tighter assessment than a lower-risk processing aid. The same chemical name does not automatically mean equivalent quality: particle size, residual solvents, microbial limits, impurity profile, polymorphic form, and packaging compatibility may all matter depending on the formulation and process.
Before requesting samples or laboratory testing, define an internal material profile. It should state the approved name, grade, pharmacopoeial or internal standard, intended function, critical quality attributes, storage conditions, retest or expiry expectations, and required release tests. This profile becomes the reference point for supplier review and incoming inspection.
A supplier assessment should go beyond commercial information. Examine whether the supplier can consistently provide the required grade and whether its documentation supports your quality system. For pharmaceutical raw materials, relevant records may include a quality questionnaire, manufacturing-site details, specification, certificate of analysis template, safety data sheet, traceability statement, change-notification commitment, and information on packaging and transport controls.
Where material criticality justifies it, a remote audit, on-site audit, or more detailed quality agreement may be appropriate. A certificate alone cannot demonstrate that a supplier’s routine controls are suitable for every critical material.

Document review often reveals problems before laboratory work begins. Compare the supplier’s certificate of analysis with the approved specification line by line. Confirm test methods, units, acceptance limits, batch number, manufacturing date, retest or expiry date, and release authorization. Results that appear acceptable may still be unsuitable when the method differs from the approved method or when an important attribute is not reported.
Incoming samples should be taken under controlled conditions by trained personnel using clean, suitable tools. The sampling plan should prevent mix-ups and contamination while providing a representative view of the batch. Check container integrity, labels, seals, shipment condition, and any evidence of temperature or moisture exposure before opening the package.
Testing should follow the approved risk-based plan. Identity testing is normally essential for each received lot unless an established and justified reduced-testing program applies. Additional testing may include assay, related substances, water content, microbial limits, residual solvents, elemental impurities, particle size, or other critical attributes. Laboratory results must be reviewed against approved limits, not merely compared with the supplier’s stated results.
Materials should remain quarantined until quality release is formally recorded. The approval record should link the supplier evaluation, specification, sample results, certificate of analysis, safety documentation, receipt inspection, and final disposition. This connection is important when a deviation, complaint, or recall requires rapid investigation.
Initial approval is not permanent. Requalification should be triggered by supplier changes, repeated incoming deviations, unexplained test variability, altered packaging, a new manufacturing site, revised test methods, or changes in regulatory expectations. A disciplined change-control process prevents an apparently minor change from entering production without assessment.
When evidence is incomplete or results conflict, do not release the batch based on schedule pressure. Place the material on hold, investigate the gap, obtain clarifying documentation or additional testing, and document the final quality decision. This approach protects both product quality and the people responsible for handling the material.
